Abstract Background The development of sensitive and specific methods for detecting Toxoplasma gondii infection is critical preventing controlling toxoplasmosis in humans other animals. Recently, various recombinant proteins have been used serological tests diagnosing toxoplasmosis. production these antigens associated with live tachyzoites obtained from cell cultures or laboratory animals genomic extraction to amplify target genes. Synthetic genes gained a key role protein production. For the first time, we demonstrated T. dense granular antigen 8 (TgGRA8) gene based on commercial synthesis. Recombinant TgGRA8 plasmids were successfully expressed an Escherichia coli system. was affinity-purified characterized via sodium dodecyl sulfate-polyacrylamide gel electrophoresis Western blotting. Furthermore, diagnostic potential assessed using 306 field serum samples goats indirect enzyme-linked immunosorbent assay (iELISA) latex agglutination test (LAT). Results blotting known positive identified single at expected molecular weight (27 kDa). iELISA illustrated that 15.40% goat -specific IgG antibodies. In addition, provided high sensitivity specificity, significant concordance (91.83) kappa values (0.69) compared results LAT. Conclusion Our findings highlight synthetic ability detect samples. specificity this could be good marker sera.
